Neural Differentiation of Human Skin Keratinocytes

Abstract

Embryonic transcription factor OCT4, is critical for maintaining cells in the undifferentiated state. We demonstrate that by transient transfection with OCT4, adult human skin keratinocytes can be reprogrammed to be more embryonic stem cell-like i.e. pluripotent. Additionally, we will illustrate through RT-PCR, Western Blot, and images of morphological changes in cell structure that these cells can be differentiated into neuronal cells with exposure to neurotrophic factors.
